37th GMP Symposium

Meet Novalix at the 37th GMP Symposium! This event is organized by the “Groupe de Métabolisme et Pharmacocinétique” (GMP), a group of researchers of the pharmaceutical industry.

The Symposium aims at promoting contact between scientists of associated disciplines, i.e. toxicologists, clinical pharmacologists, molecular biologists, cellular scientists etc.

Scientific and social programs include workshops, sessions on in vivo pharmacokinetic experiments, drug administration, new biomarkers, AI and machine learning, innovations etc.

LC-MS/MS assay for folate derivatives as proximal biomarkers of MTHFD2 inhibition in cells

We are proud to share that Dr Lionel Trottet, Head of DMPK, will present a poster on LC-MS/MS assays for folate derivatives.

Abstract: MTHFD2 is a promising target for immuno-inflammatory diseases. Understanding the mechanistic role of selective and non-selective MTHFD2 inhibitors in the THF pathway in the cellular context would be of value as this pathway is complex.

Results: Evaluation of the assay demonstrated the reliability of relative quantification of various folate derivates by LC-MS/MS. This approach allowed to show that the effect of selective vs non-selective MTHFD2 inhibitors on the THF pathway is substantially different. A notable example is the impact on 10-formyl THF directly upstream or downstream of the MTHFD1/2 enzymes. In the case of a pan MTHFD1/2 inhibitor, the cell levels of 10-formyl THF were increased up to 4-fold while in the case of selective MTHFD2 inhibitors its levels were suppressed.
